Endothelial Cell Polarization and Localized Signaling in Vascular Health

Abstract: Vascular response to blood flow forces are important for cell signaling, vessel function and overall cardiovascular health. Endothelial cells comprise the innermost lining of the vasculature and respond to these blood flow forces. Under high laminar shear stress, endothelial cells align and elongate in the direction of flow. By mapping the distribution of multiple plasma membrane associated proteins in flow aligned endothelial cells, we observe certain membrane bound proteins to polarize to one end. This polarization results in localized signaling activity that supports cardiovascular health. By manipulating endothelial cell polarization and the plasma membrane properties we are exploring how this compartmentalization of signaling events supports key biological processes within the vascular system.
